HYB 008-02-02 is specific for the Bb fragment of human factor B. Antibody binding abolishes the formation of C3bBb complexes.
HYB 008-02-02 has been successfully used in ELISA and Western blotting procedures. HYB 008-02-02 reacts strongly with factor B. Strong reaction is seen in sandwich ELISA in combination with a polyclonal antibody against factor B (e.g. DAKO A 343). In Western blotting after SDS-PAGE, HYB 008-02-02 reacts with the Bb fragment of factor B in non-reduced form only. After coupling to an activated CNBr-sepharose column, HYB 008-02-02 is suitable for affinity purification of human factor B.
The HYB 008-02-02 immunogen complement factor B isolated from human plasma.
Epitope specificity differs from that of HYB 008-06-02. Epitope specificity differs from that of HYB 008-04-02 and HAV 005-02-02 but slightly overlap as determined by inhibition ELISA.
NOTE: Concentration is lot-dependent and can vary from 0.85-1.15 mg/mL
Factor B is a component of the alternative complement pathway. The protein is a single chain polypeptide with a molecular mass of 93 kDa. Factor B is cleaved by factor D into the Ba component (30 kDa) and the Bb (63 kDa). Bb together with C3b forms the alternative pathway C3 convertase C3bBb.
